Circadian rhythms are entrained by light and influenced non-photic stimuli, such as feeding. The activity preceding scheduled mealtimes, food anticipatory (FAA), is elicited in rodents fed a limited amount at times. FAA thought to be the output of an unidentified oscillator. Previous studies, using gene deletion receptor pharmacology, implicated dopamine type 1 (D1R) signaling dorsal striatum necessary for mice. To further understand role D1R promoting FAA, we utilized Cre-lox system create cell type-specific deletions D1R, conditionally deleting GABA neurons Vgat-ires-Cre line. This conditional mutant had attenuated but was higher than expected based on prior results constitutive knockout KO Drago . result prompted us re-test original line, which expressed less controls, only moderately so. determine if genetic drift diminished effect re-established line from cryopreserved samples. reestablished Drago-cryo clear impairment compared still developed increased mealtime across 4 weeks timed Finally, tested different allele created Knockout Mouse Project.
